雌激素对Min6细胞线粒体自噬的影响

Abstract: Objective To investigate the effects of estrogen on mitochondrial autophagy in mouse islet β cells(Min6 cells). Methods Min6 cells were cultured in vitro and divided into two groups:control group(normal Min6 cells)and estrogen(E2)-treated group. The mitochondrial autophagosomes were observed using transmission electron microscopy. The expression levels of mitochondrial autophagyrelated proteins Beclin-1,HSP60,TOM20,and LC3 were detected using Western blotting. Increased expression of Beclin-1 and LC3Ⅱ/LC3Ⅰ and decreased expression of the mitochondrial matrix protein HSP60 and mitochondrial membrane protein TOM20 indicate the activation of mitochondrial autophagy. Results Formation of mitochondrial autophagosomes decreased,expression of TOM20 and HSP60 increased,and expression of Beclin-1 and LC3Ⅱ/LC3Ⅰ decreased in E2-treated Min6 cells compared with those in the control group. Conclusion Estrogen inhibits mitochondrial autophagy in Min6 cells.
