TIM-3在宫颈癌外周血T细胞中的表达及相关性研究

摘要: 目的 检测T细胞免疫球蛋白黏连蛋白-3(TIM-3)在宫颈癌患者外周血T细胞中的表达,探讨TIM-3与宫颈癌发生发展的相关性。方法 应用流式细胞仪检测CD4+T细胞,CD8+细胞及TIM-3在外周血不同类型T细胞表面的表达。收集21例对照组、15例宫颈上皮内瘤变(CIN)患者和24例子宫颈癌患者样本,进一步分析TIM-3与宫颈癌临床病理参数的关系。结果 宫颈癌及CIN患者较非宫颈病变患者外周血的CD4+T及CD8+T细胞明显减少,并且CD4+T及CD8+T细胞表面的TIM-3表达量明显增高,有统计学意义,CIN与宫颈癌相比较差异无统计学意义。与Ⅰ~Ⅱ期比较,TIM-3在宫颈癌Ⅲ~Ⅳ期外周血CD4+T及CD8+T细胞中的表达显著增加,差异有统计学意义(P<0.05)。不同临床因素分析结果显示,TIM-3在外周血T细胞中的表达与年龄、淋巴结阳性、肿瘤大小及鳞状细胞癌抗原高低无相关性(P>0.05)。结论 TIM-3与宫颈癌的发生发展有关,可能成为宫颈癌的一种新的免疫治疗靶点。

Abstract: Objective In this study,we aimed to investigate the expression of T cell immunoglobulin and mucin domain-containing protein 3(TIM-3)in T cells from the peripheral blood of cervical cancer patients,and to explore the relationship between TIM-3 and the development of cervical cancer. Methods CD4+T cells and CD8+T cells in the peripheral blood of patients with cervical cancer and CIN were significantly reduced compared with those from patients without cervical lesions,and the expression levels of TIM-3 on the surface of CD4+T and CD8+T cells were significantly increased,in a statistically significant fashion;on the other hand,no statistically significant differences were observed in samples from CIN versus cervical cancer patients. Compared with stage Ⅰ to Ⅱ,TIM-3 expression in the levels of peripheral blood of CD4+T and CD8+T cells were significantly increased in stage Ⅲ to Ⅳ cervical cancer patients(P<0.05). Analysis of different clinical factors showed that the expression of TIM-3 in peripheral blood T cells was not correlated with age,lymph node positivity,tumor size,or squamous cell carcinoma antigen level(P>0.05). Conclusion TIM-3 may affect the development and progression of cervical cancer;our data may support the use of TIM-3 for the development of novel immunotherapeutic approaches effective against cervical cancer.
