Abstract: Objective To investigate the effects of MYCT1 on the PRSS21-mediated proliferation and migration of laryngeal carcinoma cells. Methods Real-time PCR and Western blotting were used to detect PRSS21 mRNA and protein expression,respectively in the context of MYCT1 silencing or overexpression in laryngeal carcinoma. The CCK-8 and clone formation assays were applied to monitor the effects of MYCT1 on the PRSS21-mediated proliferation and survival of laryngeal cancer cells. A scratch assay was further conducted to monitor the effect of MYCT1 on the migration (PRSS21-mediated) of laryngeal cancer cells. Results PRSS21 was highly expressed in laryngeal carcinoma (P < 0.05). Compared to controls,MYCT1 significantly inhibited the expression of PRSS21 both at mRNA and protein levels in laryngeal cancer cells (P < 0.05). Importantly MYCT1 significantly inhibited the proliferation,clone formation,and migration of laryngeal carcinoma cells,in a PRSS21 dependent fashion (P < 0.05). Conclusion MYCT1 represses the proliferation and migration of laryngeal carcinoma cells via the inhibition of PRSS21 expression.
