芦丁在葡聚糖硫酸钠诱导的小鼠急性结肠炎中的作用及其机制

Abstract: Objective To establish an acute colitis model in mice using dextran sulfate sodium (DSS),and to investigate the protective effect of rutin in acute colitis and its prospective role in regulating helper T cell response in the intestinal lamina propria lymphocytes (LPL). Methods A mouse model of acute colitis was set up,where 3% DSS was used to induce colitis. The experimental group and the control group were intragastrically administrated with rutin at a dose of 50 mg·kg-1·d-1 and equal volume of normal saline,respectively. The disease activity index was calculated,and the colon histopathology score was assessed. Flow cytometry was used to detect the proportion and absolute number of helper T cell 1 (Th1) and 17 (Th17) in the intestinal LPL. Results Compared with the control group,the experimental group had slower weight loss,a lower disease activity index,milder colonic inflammation,a significantly lower histological score,and a significantly lower proportion of CD4+ and CD8+ T lymphocytes in the intestinal LPL. The proportion of interferon-γ(IFN-γ)+ cells,proportion and absolute number of interleukin-17A (IL-17A)+ cells,and absolute number of IFN-γ+ IL-17A+ cells were significantly reduced in CD4+ T cells. Conclusion Rutin plays an important protective role in the pathogenesis of DSS-induced acute colitis by inhibiting the Th1 and Th17 cell responses in the intestinal lamina propria of mice.
