长链非编码RNA RUNX1-IT1对口腔鳞状细胞癌增殖、迁移的影响及其作用机制

Abstract: Objective To understand the effect of long non-coding RNA (lncRNA) RUNX1-IT1 on proliferation and migration of oral squamous cell carcinoma (OSCC). Methods RT-qPCR was performed to evaluate the relative expression of lncRNA RUNX1-IT1 in OSCC tissues and cells. CCK8,flow cytometry and Transwell assays were performed to detect the biological functions of the OSCC cell line. Western blotting was used to measure the proteins associated with epithelial-mesenchymal transition (EMT). Results lncRNA RUNX1-IT1 expression levels were higher in OSCC tissues and cells,than those observed in adjacent normal tissues (P<0.001) and normal keratinocytes (P<0.01). Expression of lncRNA RUNX1-IT1 was significantly associated with lymph node metastasis (P<0.05). Overexpression and silencing of lncRNA RUNX1-IT1 could regulate the proteins levels of E-cadherin,N-cadherin,β-catenin,Vimentin. The expression levels of proteins involved in proliferation and migration (PCNA,Cyclin D1,and MMP2) were also upregulated or downregulated. Conclusion lncRNA RUNX1-IT1 expression levels are elevated in OSCC. We propose that lncRNA RUNX1-IT1 regulates cell proliferation and migration through EMT.
