氟伐他汀钠对皮肤鳞状细胞癌细胞增殖、凋亡及PI3K/Akt/mTOR通路的影响

摘要: 目的 探究氟伐他汀钠对皮肤鳞状细胞癌细胞增殖、凋亡及PI3K/AKT/mTOR通路的影响。方法 体外培养SCL-1细胞,MTT法检测氟伐他汀钠对SCL-1细胞增殖的影响;流式细胞术检测细胞凋亡的变化;实时qPCR法检测PI3K、Akt、mTOR mRNA相对表达量。结果 不同浓度氟伐他汀钠处理SCL-1细胞后,以浓度依赖性和时间依赖性的方式抑制SCL-1细胞增殖(P<0.01)。流式细胞术显示不同浓度氟伐他汀钠干预SCL-1细胞24 h、48 h后早期凋亡率较对照组明显升高(P<0.01),且具有浓度依赖性。不同浓度氟伐他汀钠培养SCL-1细胞24 h后PI3K、Akt、mTOR mRNA相对表达量随药物浓度的增加而降低,与对照组比较有统计学差异(P<0.01)。结论 氟伐他汀钠可抑制SCL-1细胞增殖,诱导SCL-1细胞凋亡,其作用机制可能与抑制PI3K/Akt/mTOR通路有关。

Abstract: Objective To investigate the effects of fluvastatin sodium on the proliferation,apoptosis,and PI3K/Akt/mTOR pathway in a human cutaneous squamous cell carcinoma cell line,SCL-1. Methods The MTT assay was used to detect the effect of fluvastatin sodium on the proliferation of SCL-1 cells. Flow cytometry was used to detect the apoptotic changes in SCL-1 cells after treatment with fluvastatin sodium. mRNA expression levels of PI3K,Akt,and mTOR in the cells treated with different concentrations of fluvastatin sodium were detected using RT-qPCR. Results Proliferation of SCL-1 cells was inhibited by different doses of fluvastatin sodium in concentration- and time-dependent manners (P<0.01).Flow cytometry revealed that the early apoptotic rate of SCL-1 cells treated with different concentrations of fluvastatin sodium was significantly increased after 24-h and 48-h interventions compared with the control group (P<0.01),and the apoptotic rate showed concentration dependence. The mRNA expression levels of PI3K,Akt,and mTOR in SCL-1 cells treated with different concentrations of fluvastatin sodium decreased with increase in the concentration of fluvastatin sodium at 24 h,which was statistically significant compared with the control group (P<0.01). Conclusion Fluvastatin sodium inhibits proliferation and induces apoptosis of SCL-1 cells by inhibiting the PI3K/Akt/mTOR signaling pathway.
