肌腱蛋白C在百草枯中毒肺损伤中的作用

Abstract: Objective To analyze the role of tenascin-C (TNC) in paraquat (PQ) -induced lung injury. Methods Male C57BL/6 mice (6 to 8 weeks old) were intraperitoneally injected with 30 mg/kg PQ and sacrificed on days 2,7,14,and 28 after PQ administration. We measured the expression levels of TNC protein in lung tissues of mice at all the time points when the mice were sacrificed by performing RT-qPCR,western blotting,and immunohistochemistry experiments. The protein expression levels of NF-κBp65,TGF-β1,and α-SMA in lung tissues were also measured. Results TNC protein and mRNA expression levels increased in lung tissues of C57BL/6 mice after PQ administration. Furthermore,TNC protein expression levels peaked on day 7 after PQ exposure,which was much earlier than the time point at which TGF-β1 levels were observed to have peaked. TNC was mostly detected in the extracellular matrix of the thickened alveolar septum,especially at the sites of injury. TGF-β1 showed strong expression levels during the entire duration of the experiment. NF-κBp65 showed increased expression levels only on days 2 and 7. α-SMA protein levels increased significantly during later stages of lung injury at days 14 and 28. Conclusion TNC participates in the progression of lung injury induced by PQ poisoning.
