摘要： 目的 探讨宫内发育迟缓（IUGR）大鼠胰腺组织中骨保护素（OPG）的表达水平及其对胰腺发育的影响。方法 在生后1 d及1、3、12周测量IUGR组和对照组（CON组）大鼠体质量、胰腺质量、空腹血糖及胰岛素水平，HE染色观察胰岛形态，免疫荧光双染观察胰岛β细胞增殖情况，Westen blotting及实时PCR检测胰腺中OPG蛋白和mRNA的表达。结果 与CON组相比，IUGR组大鼠胰岛体积小，β细胞增殖水平低，且在12周时出现胰岛素抵抗。IUGR组各时间点胰腺组织中OPG蛋白及mRNA表达水平均明显低于CON组。结论 IUGR大鼠胰腺中OPG蛋白及mRNA表达减少，可能导致其胰腺发育落后，胰岛β细胞增殖水平下降。

Abstract: Objective To investigate the changes in osteoprotegerin (OPG) expression levels in the pancreas of a rat model of intrauterine growth retardation (IUGR) and the effects of OPG on pancreatic development. Methods Body weight,pancreas weight,fasting blood glucose levels,and insulin levels of IUGR rats and the control (CON) group rats were measured at postnatal day 1 and weeks 1,3,and 12. Islet morphology was observed by H&E staining,islet β-cell proliferation was observed by immunofluorescence staining,and the expression levels of OPG protein and mRNA in the pancreas were detected by western blotting and real-time PCR (RT-PCR) analysis methods. Results Compared with CON group,IUGR rats displayed smaller islet volume,lower β-cell proliferation and insulin resistance after the 12-week time point. The expression levels of OPG protein and its mRNA in the pancreatic tissues of IUGR rats were significantly lower than the values obtained for CON group rats during all the time points measured. Conclusion The expression levels of OPG protein and mRNA in the pancreas of IUGR rats are decreased,which might hinder pancreatic development and decrease the proliferation levels of islet β cells in IUGR rats.
