宫内发育迟缓大鼠胰腺骨保护素的表达及其对胰腺发育的影响

Abstract: Objective To investigate the changes in osteoprotegerin (OPG) expression levels in the pancreas of a rat model of intrauterine growth retardation (IUGR) and the effects of OPG on pancreatic development. Methods Body weight,pancreas weight,fasting blood glucose levels,and insulin levels of IUGR rats and the control (CON) group rats were measured at postnatal day 1 and weeks 1,3,and 12. Islet morphology was observed by H&E staining,islet β-cell proliferation was observed by immunofluorescence staining,and the expression levels of OPG protein and mRNA in the pancreas were detected by western blotting and real-time PCR (RT-PCR) analysis methods. Results Compared with CON group,IUGR rats displayed smaller islet volume,lower β-cell proliferation and insulin resistance after the 12-week time point. The expression levels of OPG protein and its mRNA in the pancreatic tissues of IUGR rats were significantly lower than the values obtained for CON group rats during all the time points measured. Conclusion The expression levels of OPG protein and mRNA in the pancreas of IUGR rats are decreased,which might hinder pancreatic development and decrease the proliferation levels of islet β cells in IUGR rats.
