大鼠慢性肝损伤的线粒体功能障碍及其影响

Abstract: Objective To study mitochondrial dysfunction and energy abnormality in chronic liver injury in rats. Methods The chronic liver injury model was prepared by subcutaneous injection of 50% carbon tetrachloride soybean oil solution. Mitochondrial membrane potential,membrane swelling,intracellular coenzymeⅠNAD (H) content,and adenosine triphosphate (ATP) content were determined. Results Compared with the control group,the mitochondrial membrane potential level,membrane swelling degree,ATP,and coenzyme ⅠNAD (H) levels in the model group of rats were significantly decreased (all P < 0.05). The degree of injury gradually and significantly increased with time (P < 0.01). Conclusion Chronic liver injury was induced using carbon tetrachloride in rats. The observations of impaired liver mitochondrial function and abnormal energy metabolism indicate that the abnormal energy metabolism caused by liver mitochondrial dysfunction is important in diseases featuring clinical liver injury.
