阿托伐他汀对急性心肌梗死大鼠Ang-1及其受体Tie2 mRNA表达的影响

Abstract: Objective In this article,we analyzed the effect of atorvastatin treatment on the mRNA expression levels of Ang-1 and its receptor Tie2 after acute myocardial infarction (AMI) in rats and investigated the mechanism of atrovastatin activity on myocardial angiogenesis after AMI. Methods Here,80 male Sprague-Dawley (SD) rats were randomly divided into three groups:the normal group; the intervention group,which was treated with atorvastatin;and the non-drug treated intervention group. The AMI intervention group was initially administered a dose of 2.5 mg atorvastatin in solution via gavage feeding.As controls,the non-drug administered AMI group and the normal control group rats were administered with the same dosage of normal saline. Samples from all the groups were collected on 14 and 28 days after operation. The mRNA expression levels of Ang-1 and Tie2 in myocardial tissues were detected by RT-PCR. Results After 14 days of feeding,the mRNA expression levels of Tie2 in the AMI atorvastatin intervention group was significantly higher than those in the normal mice group (P < 0.05). There were no significant differences among the expression levels of Ang-1 mRNA in the intervention group,the non-drug treatment group,and the normal group (P > 0.05). Moreover,at day 28,there were no significant differences in the mRNA expression levels of Tie2 in all the three groups of rats (P > 0.05). Conclusion Atorvastatin treatment increases the expression of Tie2 mRNA and promotes the formation of new blood vessels.
