麝香通心滴丸对高血压大鼠血管内皮功能及Rho/ROCK信号通路的影响

Abstract: Objective To investigate the protective effect of musk tongxin dropping pills on the vascular endothelium in spontaneously hypertensive rats (SHR) and examine the effect on the Rho/ROCK signaling pathway. Methods SHR and normal blood pressure rats were randomly divided into normal blood pressure (control group,n=10),spontaneous hypertension model group (SHR group,n=10),and musk tongxin dropping pill groups (STX,n=10). For each animal group,the blood pressure changes were recorded. The rat serum levels of nitric oxide (NO),superoxide dismutase (SOD),and malondialdehyde (MDA) were detected using a colorimetric kit. In the blood vessels,endothelin-1 (ET-1) and vascular endothelial growth factor (VEGF) levels were detected using the enzyme-linked immunosorbent assay (ELISA);real-time PCR was used to detect the expression of ET-1 and VEGF mRNA in vascular tissues. Additionally,in the vascular tissues,the expression of RhoA,ROCK,and MLC proteins in the Rho/ROCK signaling pathway was detected using western blotting. Results Compared with the SHR group,the blood pressure of the STX group was significantly decreased. The SOD activity and NO content were significantly increased,while the MDA level was significantly decreased. The ET-1 and VEGF protein expression and mRNA levels were significantly decreased;all differences were statistically significant (P<0.05). Simultaneously,compared with the SHR group,RhoA,ROCK,and MLC protein expressions were significantly decreased in the vascular tissue of the STX group,and the difference was statistically significant (all P<0.05). Conclusion Musk tongxin dropping pills demonstrated a protective effect on the vascular endothelium of the SHR model,which could be related to the inhibition of the Rho/ROCK signaling pathway-related protein expression.
