高迁移率族蛋白B-1通过细丝蛋白A影响喉癌细胞迁移

摘要: 目的 探讨高迁移率族蛋白B-1(HMGB1)通过细丝蛋白A(FLNA)对喉癌细胞迁移能力的影响。方法 实时PCR法检测30例喉癌组织及喉癌Hep2细胞中FLNA的表达,分析FLNA的表达与患者临床病理特征的关系;RNA干扰技术敲减HMGB1表达,Western blotting和实时PCR方法检测Hep2细胞中FLNA表达;划痕实验、Transwell法检测空白对照组(CON组)、阴性对照组(NC组)、HMGB1-siRNA组、FLNA-siRNA组以及HMGB1-siRNA+FLNA-siRNA组中喉癌Hep2细胞迁移能力。结果 患者喉癌组织及喉癌Hep2细胞中FLNA低表达。FLNA表达与患者淋巴结转移、临床分期和分化程度相关(P<0.05)。与NC组比较,HMGB1-siRNA组中Hep2细胞FLNA mRNA和蛋白表达上调,细胞迁移能力显著降低(P<0.01);FLNA-siRNA组Hep2细胞迁移能力明显增加(P<0.01)。与HMGB1-siRNA组比较,HMGB1-siRNA+FLNA-siRNA组Hep2细胞的迁移能力明显降低(P<0.001)。结论 HMGB1通过抑制FLNA的表达促进喉癌细胞迁移。

Abstract: Objective To investigate the role of filamin A (FLNA) in high mobility group box-1 (HMGB1) -mediated migration of laryngeal cancer cells. Methods FLNA expression in laryngeal cancer tissues of 30 patients and in laryngeal cancer Hep2 cells was detected by real-time PCR,and the relationship between FLNA expression and clinicopathological characteristics of laryngeal cancer patients was statistically analyzed. Next,HMGB1 expression was knocked down by RNA interference and FLNA expression in Hep2 cells was detected by western blotting and real-time PCR. Wound-healing assay and transwell assay were used to detect the influence of HMGB1 and FLNA on the migration potential of Hep2 cells in CON (empty),negative control (NC),HMGB1-siRNA,FLNA-siRNA,and HMGB1-siRNA + FLNA-siRNA groups. Results FLNA expression was low in laryngeal cancer tissues and Hep2 cells,which could be related to lymph node metastasis,clinical stage,and degree of differentiation of the carcinoma (P<0.05). In HMGB1-siRNA group,the FLNA mRNA and protein expression in Hep2 cells was upregulated and the cell migration potential was significantly lower compared to the NC group (P<0.01). The migration potential of Hep2 cells in FLNA-siRNA group was significantly higher compared to that of the NC group (P<0.01),whereas the migration ability of Hep2 cells in HMGB1-siRNA + FLNA-siRNA group was significantly lower compared to that of the HMGB1-siRNA group (P<0.001). Conclusion HMGB1 promotes the migration of laryngeal cancer cells by inhibiting the expression of FLNA.
