Tenascin-c在椎间盘退行性病变中的作用及机制研究

摘要: 目的 通过观察Tenascin-c(TNC)在椎间盘退行性病变(DDD)中的表达变化,探讨TNC在椎间盘退行性病变中的作用及机制。方法 实时PCR和Western blotting检测人髓核细胞和小鼠椎间盘细胞在炎性条件下TNC、胶原蛋白Ⅱ和蛋白聚糖的变化,酶谱法及基质金属蛋白酶(MMP)活性检测MMP活性的变化。加入外源性TNC,研究TNC对DDD是否起到保护作用。体内实验验证TNC的表达情况及位置。结果 在椎间盘发生退行性病变时,TNC表达增高,胶原蛋白Ⅱ和蛋白聚糖表达降低,MMP活性增高。加入外源性TNC后,胶原蛋白Ⅱ和蛋白聚糖表达增高,MMP活性降低。结论 TNC能够调节MMP活性,对椎间盘细胞起保护作用。

Abstract: Objective To study both the effect and mechanisms of tenascin-c (TNC) on degenerative disc disease by observing changes in genetic expression of TNC in DDD. Methods We used real-time PCR and western blotting to detect the changes in TNC,CollagenⅡ,and Aggrecan in human nucleus pulposus cells and mouse intervertebral disc cells under inflammatory conditions. The changes in MMP activity were observed using zymography and MMP activity assays. Exogenous TNC supplementation was used to study whether TNC protects against DDD. In vivo experiments confirmed the expression and location of TNC. Results In the DDD,the expression of TNC was increased,collagenⅡ and aggrecan expression was decreased,and MMP activity was increased. After the addition of exogenous TNC,collagenⅡ and aggrecan expression increased and MMP activity decreased. Conclusion TNC regulates MMP activity and protects intervertebral disc cells.
