过表达ATG4A对晶状体上皮细胞上皮-间质转化的影响

摘要: 目的 探讨自噬相关基因4A(ATG4A)过表达对晶状体上皮细胞中上皮-间质转化水平的影响。方法 用细胞转染的方法在晶状体上皮细胞中过表达ATG4A并设为过表达ATG4A组(OE-ATG4A组),转染进空载体的细胞设为阴性对照组(NC组),通过实时qPCR和Western blotting方法分别检测2组细胞上皮-间质转化水平,并测定2组活性氧(ROS)水平。结果 与NC组比较,OE-ATG4A组晶状体上皮细胞上皮-间质转化水平明显升高(P<0.05),但ROS水平无统计学差异(P>0.05),说明ATG4A能够促进上皮-间质转化水平,但不影响ROS水平。结论 ATG4A能够促进晶状体上皮细胞中上皮-间质转化。

Abstract: Objective To investigate the effect of autophagy-related gene 4A(ATG4A) overexpression on epithelial-mesenchymal transformation(EMT) in lens epithelial cells. Methods An overexpression ATG4A(OE-ATG4A) group was created by transfecting ATG4A plasmid into lens epithelial cells,and a negative control(NC) group was created by transfecting a blank vector plasmid into lens epithelial cells. To analyze EMT level in both groups,quantitative reverse transcription polymerase chain reaction and Western blotting analyses were performed. The levels of reactive oxygen species(ROS) in both groups were also evaluated. Results EMT was significantly upregulated in the OE-ATG4A group compared to NC group(P<0.05);however,ROS levels did not statistically differ between the groups(P>0.05). Conclusion ATG4A can promote EMT level in lens epithelial cells.
