摘要： 目的 探讨ETV4在胃肠道间质瘤（GIST）组织中表达及其与危险度等临床病理特征的关系，以及ETV4与GIST中C-KIT，血小板衍生生长因子受体α（PDGFR-α）基因突变的关系。方法 采用免疫组化方法检测ETV4在91例GIST中的表达情况，采用直接测序法对91例GIST病例进行测序。结果 在GIST组织中，有39例（42.9%）ETV4呈强阳性表达，34例（37.4%）呈中等阳性表达。ETV4表达与患者肿瘤大小，核分裂像，Ki67增殖指数，肝转移有关（P<0.05）。ETV4表达水平与危险度呈显著正相关（r=0.387，P<0.001）；此外，ETV4表达水平与C-KIT基因突变呈显著正相关（r=0.324，P=0.008），ETV4表达水平与突变型GIST呈显著正相关（r=0.325，P=0.005）。结论 ETV4转录因子可以作为临床判定GIST生物学行为的重要指标，并且可能会成为治疗GIST的新靶点。

Abstract: Objective To investigate the ETV4 expression in gastrointestinal stromal tumors (GISTs) and its relationship with clinicopathological factors such as the risk rank of GISTs,and the relationship between ETV4 and C-KIT,a platelet-derived growth factor receptor-α (PDGFR-α) gene mutation in GISTs. Methods Immunohistochemistry was performed to assess the ETV4 expressions in 91 GISTs,for which direct sequencing was performed. Results Of the GISTs,39 (42.9%) and 34 (37.4%) showed strong and moderate positive expressions of ETV4. ETV4 expression correlated with tumor size,tumor mitotic count,the proliferation index of Ki67,and liver metastasis (P<0.05),and positively correlated with the risk rank of GISTs (r=0.387,P<0.001). In addition,it positively correlated with the C-KIT gene mutation (r=0.324,P=0.008) and the mutation type of GISTs (r=0.325,P=0.005). Conclusion The ETV4 transcription factor can be used as an important index to determine the biological behavior of GISTs and may be a new therapeutic target for GISTs.
