香菇多糖联合吉西他滨对人胰腺癌细胞ASPC-1生物学行为的影响

摘要: 目的 探讨香菇多糖联合吉西他滨对人胰腺癌细胞ASPC-1生物学行为的影响。方法 体外培养人胰腺癌细胞ASPC-1,采用MTT法检测不同浓度吉西他滨、香菇多糖单独用药以及吉西他滨和香菇多糖联合用药对细胞的增殖抑制作用,采用Annexin-V-FITC/PI双标记染色法检测对细胞凋亡的影响,采用Western blotting法检测凋亡相关蛋白survivin的表达变化。结果 吉西他滨单独用药对ASPC-1细胞的增殖抑制作用呈时间依赖性。香菇多糖单独用药在其浓度<80 μg/mL时,对ASPC-1细胞的增殖抑制作用亦呈时间依赖性,但当香菇多糖浓度达到80 μg/mL时,ASPC-1细胞对其产生耐药,增加香菇多糖的浓度抑制率无明显改变(P>0.05)。吉西他滨和香菇多糖联合用药时,对ASPC-1细胞的抑制作用明显强于二者单独用药,差异有统计学意义(P<0.05)。流式细胞术结果提示,联合用药时细胞凋亡数量较二者单独用药明显增加(P<0.05)。Western blotting结果显示,联合用药时凋亡抑制蛋白survivin表达下调。结论 体外实验中,吉西他滨和香菇多糖联合用药可协同增强对人胰腺癌细胞ASPC-1的增殖抑制作用,促进细胞凋亡,香菇多糖能对吉西他滨起到增效、增敏的效果。

Abstract: Objective To observe the inhibitory effect of lentinan combined with gemcitabine on the growth of human pancreatic cancer cell line ASPC-1. Methods ASPC-1 cells were cultured in vitro. MTT assay was used to detect the inhibitory effect of gemcitabine or lentinan monotherapy and their combined therapy on cell proliferation. Annexin-V-FITC/PI double staining was used to detect cell apoptosis. The expression of survivin was detected by Western blotting. Results The inhibitory effect of gemcitabine on the proliferation of ASPC-1 cells was time-dependent and that of lentinan was also time-dependent when its concentration was <80 μg/mL. However,when the concentration of lentinan was ≥ 80 μg/mL,ASPC-1 cells showed drug resistance,and the inhibitory rate did not change with increase in concentration (P>0.05). The inhibitory effect of the combined therapy was significantly better than that of monotherapy (P<0.05). The results of flow cytometry showed that the number of apoptotic cells in combined therapy was significantly higher than that of monotherapy (P<0.05). The results of Western blotting showed that survivin expression was downregulated when combined therapy was administered. Conclusion Collectively,the results of these in vitro experiments showed that gemcitabine and lentinan combined therapy could synergistically enhance the inhibition of ASPC-1 cell proliferation and promote cell apoptosis. Lentinan may play a synergistic or sensitizing effect in this process.
