齐墩果酸对大鼠心肌细胞缺氧后生物学功能的影响

摘要: 目的 研究齐墩果酸对缺氧大鼠心肌H9C2细胞生物学功能的影响。方法 构建缺氧H9C2细胞模型,分别用25、50 μmol/L齐墩果酸作用于缺氧的H9C2细胞,采用MTT、流式细胞术、transwell实验分别检测不同浓度齐墩果酸对H9C2细胞的增殖、凋亡和迁移的影响。采用Western blotting验证齐墩果酸对H9C2细胞p-MEK、MEK、Cyclin D1、bcl-2和MMP2的调节作用。结果 缺氧能够抑制H9C2细胞的增殖和迁移功能。25、50 μmol/L齐墩果酸能够促进H9C2细胞的增殖、抑制凋亡、促进迁移。齐墩果酸可促进p-MEK、Cyclin D1、bcl-2和MMP2蛋白的表达。结论 齐墩果酸能够通过上调p-MEK的表达,促进缺氧H9C2的生长和迁移。

Abstract: Objective To study the effect of oleanolic acid on the biological function of H9C2 cells following hypoxia. Methods The growth and migration ability of H9C2 cells were tested by performing MTT and transwell assays. The effects of 25 and 50 μmol/L oleanolic acid on the proliferation,apoptosis,and migration of H9C2 cells were studied by performing MTT,flow cytometry,and transwell analyses. Western blotting was used to verify the regulatory effect of oleanolic acid on p-MEK,MEK,cyclin D1,Bcl-2,and MMP2 in H9C2 cells. Results Hypoxia inhibited the proliferation and migration of H9C2 cells. Oleanolic acid (25 and 50 μmol/L) can promote the proliferation and migration and inhibit the apoptosis of H9C2 cells. Oleanolic acid could promote the expression of p-MEK,cyclin D1,Bcl-2,and MMP2. Conclusion Oleanolic acid can upregulate the expression of p-MEK and promote the growth and migration of H9C2 cells following hypoxia.
