Abstract: Objective To evaluate the possible therapeutic effects of Rhodiola rosea L. extracts(RhrLE)on rats' chronic restraint stress-induced anxiety,and investigate its relevant mechanisms. Methods Restraint stress was established by placing male Sprague-Dawley rats in a restraint container for 3 h/d for 21 d. RhrLE(80 and 240 mg·kg-1·d-1)were intragastrically administered to the rats 30 min before every restraint stress exposure. Twenty-four hours after the final restraint stress episode,an open field(OF)test and elevated plus maze(EPM)test were conducted to examine the anxiety-like behaviors in rats. The glutamate levels and BDNF gene expression in the hippocampus were measured using a glutamate test kit analysis and a real-time polymerase chain reaction(qPCR)assay,respectively. Results In the OF test,the restraint stress significantly reduced the total distance traveled,distance traveled in the central zone, and duration of their stay in the central zone,which were all improved by RhrLE administration. In the EPM test,chronic restraint stress significantly decreased the number of entries into the open arms and duration of staying in the open arms,while RhrLE rescued these decreases;the effects of RhrLE on the latter showed a dose-dependent relationship. Meanwhile,in the kit test and qPCR assay,RhrLE normalized the increase in glutamate levels and reduced BDNF mRNA expression in the hippocampus. Conclusion Treatment with RhrLE can ameliorate chronic restraint stress-induced anxiety in rats,which may be mediated through improving glutamatergic transmission and brain-derived neurotrophic factor system function in the hippocampus.
