Research Synopsis:
Chemistry and biology of carbohydrates. Glycobiology is the last important frontier in biochemistry. The difficulty for carbohydrate research is caused by the complexity and variety of carbohydrate structures. Carbohydrate molecules become the biomolecules carrying the largest amounts of bio-information. As information carriers, carbohydrates play essential roles in living organisms, including determination of antigenecity of molecules and phenotype of cell, and recognition in many physiological and pathological events. The simulative syntheses of the bioactive oligosaccahrides for verifying important conclusions of bio-functions of natural oligosaccharides and further chemical modification for obtaining more active non-natural oligosaccahrides are very important for study on structure-bio-function relationships.
Gene delivery system Thekeytechnologyforgenetherapyistodesignaneffectiveandsafevectorfortargetedgenedelivery.Cationicliposomebasednon-viralgenevectorsaredesigned,synthesizedandevaluatedbyinvitrotransfection.Thetransfectionpropertiesandtargetedassayofthosegenevectorswillbediscoveredbyobservingenhancedgreenfluorescentprotein(EGFP)expressionfrompEGFP-C1plasmidinHEK293,293FT,GES-1,GC-1,SW-480,HT-1080,U251,3T3-L1,MCF-7,Hct-116,HUVEC,HeLa,A549,HBL100-SV40andNIH3T3cells.Andastructure-activityrelationshipbasedonthechemicalstructureandphysicalstructureoflipidsgenedeliveryvectorswillbeestablishedbyevaluatingtheeffectsofthechemicalstructureandphysicalstructuresuchaslengthofhydrophobicchain,lengthofthelinkagebetweenhydrophilicterminalandhydrophobicterminal,hydrophilicterminaltypes,hydrophilicterminaldensity,surfacezetapotential,particlesizeandsizedistribution,onthetransfectionproperties.Thisstructure–activityrelationshipwillbetoprovideaguidingtheoryforconstructingnewcationicliposome-basednon-viraltargetedgenedeliveryvectorsandtheirapplication.
