Dr. Liangyi Chen earned his B.S. in Biomedical Engineering from Xi’an Jiaotong University in 1995. He then joined Prof. Huaguang Kang’s lab in Huazhong University of Science and Technology and got his Ph.D in Biophysics and Biochemistry in 2001. Then Dr. Chen did his postdoctoral study in Prof. Bertil Hille’s lab at University of Washington, Seattle, WA from 2001 to 2004. From 2004 to 2010, he was an associate investigator in the Institute of Biophysics, Chinese Academy of Sciences. From 2010 till now, he is a professor and principal investigator in Institute of Molecular Medicine, Peking University.
His research interests are focused on development of state-of-the-art high spatiotemporal resolution fluorescence imaging techniques (including optics and algorithms development) for in vivo studies. His group also uses these new technologies to study the mechanism of blood glucose regulation and the pathogenesis of diabetes, using zebrafish and mouse models. As the corresponding author, Dr. Liangyi Chen has published papers in peer-reviewed journals such as Dev Cell, Cell Res., Proc Natl Acad Sci U S A., Biophys J., Biomed Opt Express., Diabetes and Diabetologia, etc. He has been servingas a senior faculty member in the Neuronal Signaling Mechanisms Section in Faculty of 1000 Biology since 2012. He has been invited to give talks in international conferences organized by academic societies such as OSA, SPIE and Biophysical Society, etc.
In summary, his group focuses on the following aspects:
1. Development of state-of-the-art live cell imaging technologies.
1) 2011 Four-color total internal reflection fluorescence microscope (TIRF)
2) 2013 Super-axial-resolution scanning TIRF microscope (va-TIRF), 50 nm Z axial resolution (Biomed Opt Express, 2014)
3) 2014 Two-photon three-axis digital scanned light-sheet microscope (2P3A-DLSM, Cell Res., 2015)
4) 2014 Automated fluorescence images analysis and vesicle fusion determination platform (Biophys J. 2015)
5) 2015 Super-resolution structure illumination microscope (Super-SIM, 90 nm spatial resolution) for live cell imaging, pending publication
6) 2016 unsupervised machine learning methods for accurate phenotyping of large fluorescence image datasets
2. Mechanisms of Exo-endocytosis coupling in pancreatic beta-cells.
1) Hopping of Clathrin-coated pits as a new mechanism for the exocytosis-coupled endocytosis (Dev Cell, 2015)
2) Number of SNARE complexes required for different types of vesicle exocytosis
3) Mechanisms of the fast, clathrin-independent endocytosis in pancreatic beta-cells
