研究领域近20年,爆发了一些可同时感染人和牲畜的新生、再生病毒性流行病,其中很大一部分是由吸血的节肢动物叮咬脊椎动物造成病毒(虫媒病毒)感染引起的。虫媒病毒分为黄病毒科(Flaviviridae)和披膜病毒科(Togaviridae)等。登革热病毒(Dengue Virus)就是由蚊虫传播的黄病毒科的虫媒病毒。最近的研究显示,每年约有4亿人感染登革热病毒,还有25亿人处于接触登革热病毒的高风险之中。大部分的登革热病毒感染表现为轻症并可以自愈,但仍有部分病例会发展成严重的登革出血热(DHF)/登革休克综合症(DSS)甚至死亡。另一种重要的虫媒病毒是披膜病毒科、甲病毒属(Alphavirus) 的基孔肯雅病毒(Chikungunya Virus)。从2005年起,基孔肯雅病毒通过新的载体,白纹伊蚊,扩大了其传播范围,甚至传播到了加勒比海地区和美洲大陆。这两种虫媒病毒给公共卫生和安全带来了巨大的威胁,但目前还没有针对性的疫苗和药物,关于这些病毒在细胞和分子水平上的细胞嗜性和致病机理更是存在很多未知。我们以这两种虫媒病毒为研究对象,感染哺乳动物细胞和蚊虫,通过体内外实验,揭示病毒致病因子、宿主促病毒因子或者限制性因子。特别关注i)在病毒进入细胞、跨种传播和易感性中发挥重要作用的宿主因子ii)蚊虫体内影响病毒复制和传播的细胞反应和信号通路。这些研究将有利于研发防治虫媒病毒的新策略。
