一、病毒学研究
1)H5N1型禽流感病毒致病机制的研究
进行禽流感NS1、NS2基因与宿主细胞大分子相互作用研究,以揭示引起不同种属细胞炎性因子激活关键分子的过程及相互作用机制。对禽流感病毒感染细胞后NS1基因在细胞内聚集的相关生物学特征,NS2基因在不同细胞中对重要基因调节的信号网络的研究。
2)基于小环DNA载体的病毒疫苗研究
利用小环的低免疫原性及安全性,进行非病毒载体小环DNA的HIV、HPV等病毒疫苗方面的研究。并对其在真核细胞表达特性,免疫原性及受体-配体靶向性等方面进行研究。
3)EBV病毒致瘤机制及疫苗方面的研究
研究EBV核抗原EBNA1蛋白参与p53途径调节的机制。另外,鉴于EBNA1广泛表达于各种EBV相关的肿瘤中,研究更为安全有效的针对加强CTL反应的疫苗。
4)ADAR1的抗病毒机理研究
ADAR1在细胞内可以特异性的编辑双链RNA分子,从而产生抗病毒效应。研究选用流感病毒、狂犬病毒等对人类社会危害较大的RNA病毒作为研究对象,主要研究细胞内ADAR1的表达量是否会影响RNA病毒在宿主细胞内的复制作用,并深入探讨ADAR1的抗病毒机理。
二、肿瘤学研究
1)靶向性纳米材料介导的抗肿瘤研究
药物纳米载体具有高度靶向、药物控制释放、提高难溶药物的溶解率和吸收率等优点,因此我们致力于构建智能性纳米,使其能够定向高效的杀伤特定的肿瘤细胞,减少对非特异组织的杀伤。
2)受体酪氨酸激酶抑制剂的抗肿瘤研究酪氨酸激酶抑制剂(TKIs)的抗肿瘤作用机制是通过以抑制肿瘤细胞的损伤修复、使细胞分裂阻滞在G1期、诱导和维持细胞凋亡、抗新生血管形成等途径实现的。TKIs还可通过下调肿瘤细胞的血管生成因子以及抑制EGFR对肿瘤血管内皮细胞的信号传导,EGFR和血管内皮生长因子受体(VEGFR)两种信号传导通路的“交叉对话”,为临床同时抑制这两种传导通路提供了合理的依据。
